3.3 Method Of The Research
Process of the research that are:
1.Preparation of tools and materials to do the research
2.Take matured female dumbo catfish for weighing
3.Put the female in the basin
4.Take donor fish (male and female carp) and weighing it. The weight of donor are balance with the dumbo broodstock, because this research is using dose about 1:1
5.Cut the head of carp and cleave the upper side (above its nose) and take its hypophysa
6.Place it in alcohol in order to poure out blood from hypophysa
7.Move hypophysa into mortar and put aquabidest into it
8.Smash the hypophysa until it run
9.Take the extract of hypophysa gland with spuit and moved into centrifuge
10.Turn around the centrifuge for several minutes until homogenous dilution is created
11.Take again the dilution with clean spuit
12.Inject it to the back part with slope 450 with depth is 2 cm
13.Input the injected broodstock into the basin. Male and female are palced in separate basin.
14.Take egg sample out every 2 hours to be observed under microscope (observation about egg development before it’s been strriped out).
